If you mean medications that can significantly worsen cognition or increase dementia risk, there isn’t a medically valid list of seven drugs that cause “severe dementia and profound brain damage” in everyone who takes them. The risk depends heavily on dose, duration, age, other medications, and underlying illness.
Medications/classes that deserve particular attention include:
Diphenhydramine (Benadryl) — a strong anticholinergic; can cause confusion and delirium, particularly in older adults.
Oxybutynin — anticholinergic used for overactive bladder; higher anticholinergic burden is associated with cognitive impairment.
Amitriptyline — tricyclic antidepressant with substantial anticholinergic effects.
Benzodiazepines such as lorazepam, alprazolam, clonazepam, and diazepam — can cause sedation, memory problems and cognitive impairment; long-term use is particularly concerning.

Important: Don’t abruptly stop benzodiazepines or other medicines you’ve been taking regularly. Benzodiazepine withdrawal can cause seizures and other potentially life-threatening complications, so they should generally be tapered under medical supervision.
